Se caracteriza por la acumulaci\u00f3n de una prote\u00edna, la alfa-sinucle\u00edna, en las neuronas dopamin\u00e9rgicas de la sustancia nigra y en otras \u00e1reas del sistema nervioso.<\/p>\n<blockquote><p><strong><em>La administraci\u00f3n en ratones de este oligonucle\u00f3tido conduce a la reducci\u00f3n de la expresi\u00f3n del gen de la alfa-sinucle\u00edna en un tipo de neuronas concretas<\/em><\/strong><\/p><\/blockquote>\n<p>Esto provoca alteraciones de la funci\u00f3n del circuito dopamin\u00e9rgico y la presencia de s\u00edntomas motores como temblores, lentitud de movimientos, as\u00ed como tambi\u00e9n diversos s\u00edntomas no motores como el trastorno del sue\u00f1o en fase REM, disfunciones olfativas, depresi\u00f3n, deficiencias cognitivas y demencia. Los tratamientos actuales son sintom\u00e1ticos y la mayor\u00eda modulan la actividad de los circuitos motores.<\/p>\n<p>Para el estudio se ha utilizado un oligonucle\u00f3tido inhibitorio, ASO (antisense oligonucleotide), dirigido contra la alfa-sinucle\u00edna para inhibir su s\u00edntesis. Los oligonucle\u00f3tidos son mol\u00e9culas formadas por secuencias cortas de ADN o ARN que se utilizan en terapia g\u00e9nica como estrategia para el silenciamiento o inhibici\u00f3n de genes.<\/p>\n<p><strong>Optimizar la neurotransmisi\u00f3n<\/strong><\/p>\n<p>Los investigadores han demostrado que la administraci\u00f3n por v\u00eda intranasal en ratones de este oligonucle\u00f3tido permite su acumulaci\u00f3n en el cerebro y conduce a la reducci\u00f3n de la expresi\u00f3n del gen de la alfa-sinucle\u00edna en un tipo de neuronas concretas.
