Este m\u00e9todo, que simplifica y mejora la obtenci\u00f3n de im\u00e1genes de las mol\u00e9culas de la vida, ha llevado a la bioqu\u00edmica a una nueva era.<\/p>\n<p>Las im\u00e1genes son claves para entender cualquier proceso. Los descubrimientos cient\u00edficos a menudo se basan en poder visualizar bien los objetos que son invisibles al ojo humano. Sin embargo, hasta la fecha los &#8216;mapas&#8217; bioqu\u00edmicos se han llenado de espacios en blanco porque la tecnolog\u00eda disponible ten\u00eda dificultades para generar im\u00e1genes de gran parte de la maquinaria molecular de la vida.<\/p>\n<p>Pero ahora la llamada criomicroscopia electr\u00f3nica lo cambia todo. Los cient\u00edficos pueden congelar las biomol\u00e9culas en pleno movimiento y visualizar procesos que nunca antes se hab\u00edan visto, lo que es decisivo tanto para la comprensi\u00f3n b\u00e1sica de la qu\u00edmica de la vida como para el desarrollo de productos farmac\u00e9uticos.<\/p>\n<p>Durante mucho tiempo se pens\u00f3 que los microscopios electr\u00f3nicos solo eran adecuados para captar im\u00e1genes de materia muerta, ya que el poderoso haz de electrones que emplea destruye el material biol\u00f3gico. Pero en 1990, el escoc\u00e9s Richard Henderson (Edimburgo, 1945) logr\u00f3 por fin utilizar uno de estos microscopios para generar una imagen tridimensional de una prote\u00edna a resoluci\u00f3n at\u00f3mica. Este avance demostr\u00f3 el potencial de la t\u00e9cnica.<\/p>\n<p><strong>Fusi\u00f3n de im\u00e1genes difusas 2D para crear una n\u00edtida 3D<\/strong><\/p>\n<p>Por su parte, el qu\u00edmico alem\u00e1n Joachim Frank (Siegen, 1940) hizo que esta tecnolog\u00eda se pudiera aplicar de forma general. Entre 1975 y 1986 desarroll\u00f3 un m\u00e9todo de procesamiento de im\u00e1genes en el que se analizan las fotograf\u00edas bidimensionales difusas que toma el microscopio electr\u00f3nico y despu\u00e9s se fusionan para revelar una estructura tridimensional definida.<\/p>\n<blockquote><p><strong><em>La resoluci\u00f3n at\u00f3mica que ansiaban los cient\u00edficos se alcanz\u00f3 en 2013 y hoy se producen de forma rutinaria las estructuras 3D de las biomol\u00e9culas<\/em><\/strong><\/p><\/blockquote>\n<p>Y la aportaci\u00f3n del suizo Jacques Dubochet (Aigle, 1942) fue el uso del agua en la microscop\u00eda electr\u00f3nica. El agua l\u00edquida se evapora en el vac\u00edo que se genera dentro de estos avanzados microscopios, lo que hace que las biomol\u00e9culas colapsen. Sin embargo, a principios de los a\u00f1os ochenta, Dubochet consigui\u00f3 vitrificar el agua: la enfri\u00f3 con tanta rapidez que se solidific\u00f3 en su forma l\u00edquida alrededor de una muestra biol\u00f3gica, permitiendo as\u00ed que las biomol\u00e9culas conserven su forma natural, incluso en el vac\u00edo.<\/p>\n<p>Despu\u00e9s de estos descubrimientos, se han optimizado todas las piezas del microscopio electr\u00f3nico. La resoluci\u00f3n at\u00f3mica que ansiaban los cient\u00edficos se alcanz\u00f3 en 2013, y ahora se pueden producir de forma rutinaria las estructuras tridimensionales de las biomol\u00e9culas.<\/p>\n<p>En los \u00faltimos a\u00f1os, la literatura cient\u00edfica se ha llenado de im\u00e1genes de todo tipo, desde prote\u00ednas que causan resistencia a antibi\u00f3ticos hasta la superficie del virus Zika.
